How have you connected CTP_RST? That would be TouchPanel ReSeT. It won't respond if it's in reset.
Hold on I'm not sure I understand what's causing what. Is i2cdetect not detecting the screen because the overlay/driver isn't right, or is the overlay/driver not initializing correctly because the screen isn't being detected?
I2cdetect is just doing a low level i2c probe. If you haven't brought the chip out of reset then it won't respond.
How you control the reset line is totally up to you, either through an overlay (once you have everything working), manual gpio controls, or using hardware pull up/downs.
